He attended William Jewell College and the University of Missouri, where he served as the first student body president in 1911. Following his service as a captain in the Army Engineer Corps during World War I, he co-founded Carter-Waters in 1922 with Albert R. Waters, a construction materials firm that remains in operation.

E. Kemper Carter married Anna Curry on Washington's Birthday in 1936.
